At Gotoku Chemical Co., Ltd., around 1955, the concept of bioavailability began to spread throughout the pharmaceutical industry in Europe and the United States, and along with this the need for drug elution time grew. In order to put this into practice, the disintegration of granules and tablets is important, and disintegrants have been developed in Europe and the United States. Around 1970, starch-based Primojel, cellulose-based Ac-Di-Sol, and organic polymer-based Polyplasdone XL were born as famous superdisinternts. The characteristic of these three types is that they all have a cross-linked structure. In the 1960s, trivet medicines had a strong interest in cellulose derivatives, and regardless of Europe and the United States, they succeeded in developing a disintegrant that reacted with the substituent R-COOH by using the chelating action of Ca, although it was not as strong as cross-linking. We have developed a three-dimensional structure. That is the ECG-505.

Since then, the market has grown to the point where it can fully compete with Europe and the United States. Free CMC, which plays a role as a raw material in the production of ECG-505, is not suitable as a disintegrant for swelling because the substituents R-COOH and hydroxyl groups are aggregates in an acid form that does not absorb water well and has a weak swelling power. It was something, subsequent research revealed that it was a wicking type disintegrant, and NS-300 was developed as a new disintegrant. Under these circumstances, Gotoku Chemical has succeeded in creating two types of cellulose-based disintegrants with different purposes, swelling, and wicking which have continued to this day.

The demand for bioavailability increased in the pharmaceutical industry, which was attributed to the rapid release of drugs. Gotoku Chemical also used its experience with CMC-Na to successfully develop a unique CMC-Ca (carmellose calcium) ECG-505, followed by CMC-H (carmellose) NS-300 is extensively used in the pharmaceutical industry. ECG-505 was manufactured in 1962, and NS-300 was manufactured by Nichirin Chemical Industry Co., Ltd., in 1967 which has a GMP system. In particular, ECG-505 is a swelling type, and NS-300 is a wicking type.
